- Chief Jumacao was the first Taíno Chief to learn how to read and write. This fact was corroborated when he wrote a letter to the King of Spain complaining about the Governor of the island not complying with the peace treaty, and that other chiefs as well as himself were virtually prisoners of the regime. He stated also in this communication that he was a free man and capable of responding for his own acts. The King was so moved by the letter, that he ordered immediately that Puerto Rico honor the terms of the peace treaty.
- Humacao's people nickname is "los roye huesos"(the bones grinders).
- Some people says that the gray view of the city was due the smoke of the sugar cane centrals.
- Off shore near Humacao is an island inhabited by Rhesus monkeys once used for scientific experiments back in the 1930s. Cayo Santiago (38 acres) host one of two field stations of the Caribbean Primate Research Center in the island.
- Humacao is famous all over Puerto Rico for its granos, a greasy snack food which is composed of fried rice flour with cheese in its center. People have for years driven to Humacao from other cities just to taste this snack food sell in little carts all over the downtown. Locals joke about this tasteful 'fritura': el granito es EEE: engorda, estriñe y embrutece.This means that the "granos" makes you fat, constipate you, and turns you stupid.
- On March, 2008, a new Roman Catholic diocese was established as the Fajardo-Humacao diocese. Its first bishop is Monsignor Eusebio 'Chebito' Ramos Morales, a maunabeño who was rector of the Humacao's main parish in the 1990's.
